Cytotoxicity and variant cellular internalization behavior of water-soluble sulfonated nanographene sheets in liver cancer cells

چکیده
Highly exfoliated sulfonated graphene sheets (SGSs), an alternative to graphene oxide and graphene derivatives, were synthesized, characterized, and applied to liver cancer cells in vitro. Cytotoxicity profiles were obtained using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide, WST-1[2-(4-iodophenyl)-3-(4-nitrophenyl)-5-(2,4-disulfophenyl)-2H-tetrazolium, and lactate dehydrogenase release colorimetric assays. These particles were found to be non-toxic across the concentration range of 0.1 to 10 μg/ml. Internalization of SGSs was also studied by means of optical and electron microscopy. Although not conclusive, high-resolution transmission and scanning electron microscopy revealed variant internalization behaviors where some of the SGS became folded and compartmentalized into tight bundles within cellular organelles. The ability for liver cancer cells to internalize, fold, and compartmentalize graphene structures is a phenomenon not previously documented for graphene cell biology and should be further investigated.

Optimized Cytoplasmic Expression of Water Soluble Human Thrombopoietin in Modified Bacterial Strain
Background: Thrombopoietin is a glycoprotein produced by liver and kidney which is responsible for regulating the platelet production. Thrombopoietin is a key ligand with impact on regulating the self-renewal of Hematopoietic stem cells and the regulation of Megakaryocytes progenitors. Previous studies have indicated that only N-terminal domain of this protein has receptor promoting ability. Th...
متن کاملSynthesis, characterization and cytotoxicity of alkylated quercetin derivatives
Quercetin, a ubiquitous flavonol, represents a promising leading drug for development of new chemotherapeutic agents. However, its limited cytotoxicity to cancer cells hampers its clinical use. In order to obtain novel quercetin derivatives with superior cytotoxicity, seven alkylated quercetin derivatives were synthesized. Solubility of these derivatives was determined by turbidimetry. Cytotoxi...

Introduction: In recent years, bismuth-based nanomaterials have been widely used in medical researches as imaging, drug delivery and x-ray radiosensitizing agents. Due to their anti-microbial effects against Helicobacter pylori (HP), bismuth colloidal compounds are used to treat various types of diseases such as chronic gastritis. Despite their advantages, bismuth-based compoun...
متن کاملDifferential Attachment of Pulmonary Cells on PDMS Substrate with Varied Features
Cancer is now a global concern, and control of the function of cancer cells is recognized as an important challenge. Although many aggressive chemical and radiation methods are in practice to eliminate cancer cells, most imply severe adverse toxic effects on patients. Taking advantage of natural physical differences between cancer and normal cells might benefit the patient with more specific cy...
متن کاملEVALUATION OF ANTI-CANCER ACTIVITY OF COVALENTLY CONJUGATED METHOTREXATE TO POLYAMIDOAMINE GENERATION 4 DENDRIMER ON MCF-7 CANCER CELLS: AN EXPERIMENTAL STUDY
Background & Aims: Poly (amidoamine) dendrimer (PAMAM) is highly macromolecular at nanosize with widely active amine groups on the surface that allows it to attach to the anti-cancer drugs such as Methotrexate (MTX). This study aimed to synthesize and characterize PAMAM-MTX (dendrimer-MTX) complex, then to evaluate the cytotoxic effect of the synthesized complex on MCF-7 cancer cells. Material...
